Study: Investigating the role of Glut1 in the microvascular complications of diabetes

Nearly 25% of veterans are affected by diabetes. Diabetic microvascular complications occur in up to 60% of diabetic patients, significantly reducing quality of life and increasing mortality. Diabetic Retinopathy is the leading cause of blindness, Diabetic Kidney Disease is the leading cause of end-stage renal disease and Diabetic Peripheral Neuropathy is the leading cause of non-traumatic lower extremity amputation. However, few effective therapies to slow or prevent the development of each disorder are currently prescribed. While each comorbidity develops independently, hyperglycemia is a major risk factor for all, and intensive glycemic control mitigates their severities. Because glucose levels in the eye, kidney and peripheral nerves approximate blood glucose levels, systemic hyperglycemia leads to chronic oxidative stress, inflammation and damage in each tissue. This damage in turn compromises microvascular function and drives diabetic eye, kidney and peripheral nerve pathology. Therefore, it is important to identify ways to prevent these complications.
Our research aims to determine (1) whether systemic reduction of the facilitative glucose transporter, Glut1, prevents these complications by lowering glucose levels in each tissue, and (2) to elucidate molecular mechanisms that regulate Glut1 in each tissue. Our goal is to determine whether reduced glucose transport via Glut1 is effective in preventing onset of each of the microvascular complications of diabetes and to identify novel therapeutic targets for reduction of glucose transport into compromised tissues.

Compliance Office Tip of the Month:
When consenting a subject, remember to always thoroughly check the documents for completeness and accuracy – before the subject leaves the room! This helps ensure you’ve covered everything, helps to check again that the subject has understood everything presented to them, and it gives you an opportunity to catch mistakes while they are still easily fixable! Come prepared to consenting appointments with pens that work well and a checklist to help yourself remember all aspects of the consenting process. Finally, don’t forget to provide the subject a complete copy of all consenting documents that they sign.

GRECC Statistical Resource Center (SRC)
The GRECC SRC team is a valuable resource for researchers, offering a range of services to support and advance your research endeavors.
Here are some of the services provided by the SRC:
- Study design
- Assistance with IRB study submission (analytic plan only)
- Support with grant preparation (analytic plan only)
- Guidance on planning data acquisition -
- DSMB reports
- Data analysis
- Data visualization
- Help with methods documentation in manuscripts/presentations
Researchers can reach out to the GRECC SRC team for assistance with statistical inquiries, study planning, or data analysis. No funding is required to access their services.

Study Initiation Visits
What is a Study Initiation Visit?
The study initiation visit encompasses a review of regulatory requirements as they apply to your specific study, good clinical practice guidelines, the informed consent and HIPAA documents as well as a review of how to maintain study records (regulatory binder and subject files).
The Research Compliance Office is required to perform a triennial regulatory audit of studies (human subjects' research, animal protocols, and basic science studies) and informed consent and HIPAA audits if applicable. The intent of a study initiation visit is to help the researcher avoid the most common audit findings before they occur.
